I have never been to Ding Darling but from what I have heard, there is more to see because you are directly on the water. There is a toll to get to Sanibel from Fort Myers so once you are there, you can explore the island and Captiva. At Six Mile Cypress you are walking on a boardwalk whereas at Ding Darling, you can either drive through, stopping along the way, or bike. There are bike rentals. Sanibel is busy during "season" so be prepared for crowds.
I want to visit the Six Mile Slough Preserve and would like to stay at a beachfront resort as close to Preserve as possible. I prefer resorts without carpet. Any advice?
